چکیده
Chloride intracellular channel (CLIC) is a member of the chloride protein family for which growing evidence supports pivotal role in fundamental cellular events. However, physiological function CLIC insects still rarely uncovered. The ovary-derived High Five (Hi-5) cell line isolated from cabbage looper ( Trichoplusia ni ) widely used laboratories. Here, we studied both characteristics and functions Hi-5 cells Tn CLIC). We identified TnCLIC gene annotated highly conserved proteins most insect species. After RNA interference , phenomenon significantly increased death suggests that essential survival cells. same lethal effect was also observed Spodoptera frugiperda 9 Drosophila melanogaster Schneider 2 after knockdown. Furthermore, found this kind accompanied by increases calcium ions knockdown with transcriptomic analyses detection levels. Our results provide insights into as key factor lay foundation mechanism.

Live cell imaging using confocal microscopy induces intracellular calcium transients and cell death.
Isolated chondrocytes stained with fluo 4-AM and visualized using standard confocal microscopy techniques exhibited Ca2- transients and oscillations. Decreasing the power of the laser light decreased the percent-age of cells exhibiting these Ca2+ signals. Treatment with the antioxidant ascorbate reduced the Ca2+ response, suggesting that it was mediated by light-induced release of reactive oxyg...
متن کاملAntagonists of the TMEM16A calcium-activated chloride channel modulate airway smooth muscle tone and intracellular calcium.
BACKGROUND Perioperative bronchospasm refractory to β agonists continues to challenge anesthesiologists and intensivists. The TMEM16A calcium-activated chloride channel modulates airway smooth muscle (ASM) contraction. The authors hypothesized that TMEM16A antagonists would relax ASM contraction by modulating membrane potential and calcium flux. METHODS Human ASM, guinea pig tracheal rings, o...
متن کاملCell swelling increases intracellular calcium in Necturus erythrocytes.
This study examined the role of Ca(2+) in regulatory volume decrease by Necturus erythrocytes. Hypotonic shock (50% tonicity) stimulated an increase in cytosolic free Ca(2+), detected using epi-fluorescence microscopy and the fluorescent Ca(2+) indicator fluo-4-AM (10 microM). A similar increase in fluorescence did not occur under isosmotic conditions, unless cells were exposed to the Ca(2+) io...
متن کاملEvidence for chloride ions as intracellular messenger substances in astrocytes.
Cultured rat hippocampal astrocytes were used to investigate the mechanism underlying the suppression of Ba2+-sensitive K+ currents by GABAA receptor activation. Muscimol application had two effects on whole cell currents: opening of the well-known Cl- channel of the GABAA receptor and a secondary longer-lasting blockade of outward K+ currents displaying both peak and plateau phases. This block...
متن کاملInfrasound increases intracellular calcium concentration and induces apoptosis in hippocampi of adult rats.
In the present study, we determined the effect of infrasonic exposure on apoptosis and intracellular free Ca²⁺ ([Ca²⁺]i) levels in the hippocampus of adult rats. Adult rats were randomly divided into the control and infrasound exposure groups. For infrasound treatment, animals received infrasonic exposure at 90 (8 Hz) or 130 dB (8 Hz) for 2 h per day. Hippocampi were dissected, and isolated hip...
